If somebody signed a handwritten paper saying that they agree to a settlement, can they change they're mind?

A settlement is a contract.  It is valid if the elements of a contract are met, and handwritten contracts are valid.  If you want to change your mind on a settlement, then you need to consult a lawyer immediately.  There may be a valid reason to avoid the settlement, but time will only make this more difficult.
